Standard T Cell Processing + New ReagentTo make CaspaCIDe™ DLI, T cells are isolated from the donor's peripheral blood and transduced with the CaspaCIDe™ vector. The vector includes a ΔCD19 marker which is used to select cells that have been successfully transduced, ensuring the final CaspaCIDe™ DLI product consists of high purity, AP1903-responsive CD3+CD19+ T cells. In the ongoing CASPALLO clinical trial, donor T cells are also allodepleted prior to transduction, to minimize the risk of GvHD. This complex step was incorporated as a precaution, before the effectiveness of CaspaCIDe™ had been established. This non-essential processing step will be omitted in future studies. Cell processing can be done locally, at any GMP/GTP compliant transplant center with suitable cell processing capabilities, or at a central manufacturing facility. |
